چکیده

A quantitative empirical model is presented, which relates the total surfactant concentration with the fluorescence intensity and the mean translational diffusion coefficient of dyes in micellar solutions, as determined by Fluorescence Correlation Spectroscopy (FCS). Based on this model a systematic data analysis method is defined for the precise determination of the dye binding equilibrium constant, the critical micelle concentration, the translational diffusion coefficients, and the hydrodynamic radii of dyes 10

Unfolding dynamics of cytochrome c revealed by single-molecule and ensemble-averaged spectroscopy.

Denaturant-induced conformational change of yeast iso-1-cytochrome c (Cytc) has been comprehensively investigated in the single-molecule and bulk phases. By fluorescence-quenching experiments with dye-labelled heme-protein (Alexa 488-labelled Cytc, Cytc-A488), we clearly show that the fluorescence quenching observed from folded Cytc-A488 is due mainly to photoinduced electron transfer (PET) bet...

Ensemble and Single-Molecule Studies on Fluorescence Quenching in Transition Metal Bipyridine-Complexes

Beyond their use in analytical chemistry fluorescent probes continuously gain importance because of recent applications of single-molecule fluorescence spectroscopy to monitor elementary reaction steps. In this context, we characterized quenching of a fluorescent probe by different metal ions with fluorescence spectroscopy in the bulk and at the single-molecule level. We apply a quantitative mo...

The Effect of dye-dye interactions on the spatial resolution of single-molecule FRET measurements in nucleic acids.

We study the effect of dye-dye interactions in labeled double-stranded DNA molecules on the Förster resonance energy transfer (FRET) efficiency at the single-molecule level. An extensive analysis of internally labeled double-stranded DNA molecules in bulk and at the single-molecule level reveals that donor-acceptor absolute distances can be reliably extracted down to approximately 3-nm separati...

Influence of a Crown Ether on the Spectral Properties of Triarylmethane Dyes in Aqueous and Micellar Environments

This study demonstrates the spectral data for two triarylmethane dyes, malachite green (MG) and crystal violet (CV) at room temperature. Effects of micellar environment, and crown ether addition on the spectral behaviour of the dyes were studied using absorption spectroscopic method. The dye-anionic surfactant interactions in micellar media were investigated. The chosen surfactants with differe...

Acid-base behavior of rhodamine B in a reversed micellar medium of cetyltrimethylammonium chloride in 1-hexanol-cyclohexane/water.

The acid-base behavior of rhodamine B (RB) in reversed micellar solutions of cetyltrimethylammonium chloride (CTAC) in 1-hexanol-cyclohexane/water was investigated through absorption and fluorescence measurements under varying the hydrogen-ion concentration in a water pool, the mole fraction of 1-hexanol in a bulk solvent and the water to surfactant molar concentration ratio. RB exhibited equil...
